Effect of pH of Culture Medium on Results of Macrobroth Dilution Antifungal Susceptibility Testing of Candida albicans


Abstract
Mycotic infection seems to show a increasing tendency in recent years because the incidence of cancer and transplantation is increased, and their frequent complication is also encountered.
Particularly, C. albicans infection can cause a serious damage in immunosuppressed patient if systemically disseminated.
Thus, the antifungal susceptibilty testing may be essential, but not yet clearly established, that reason is why several factors for such a test exists and among these, the inoculum dose, culture duration and meduim condition (pH, component) can be noted.
The present study was designed to investigate the effect of pH of culture medium on results of macrobroth dilution antifungal susceptibility testing of C. albicans.
The results were obtained as follows:
1. the change of culture medium gave a significant change in MIC value of Ketoconazole (KTZ).
2. in case of buffered SDB medium, the value of MIC remained stable compared to that of unbuffered medium through all culture time.
3. about pH 7.0 in buffered SDB showed low and narrow range of MIC value of KTZ.
4. the change of MIC value under each pH of buffered SDB was almost same as the expected MIC value which is calculated on the basis of piperazine protonation rate.
In conclusion, the pH of the SDB gabe a significnat change in the MIC value of KTZ. this phenomena can be explained by the change of piperazine protonation rate under the gaven pH.
